From the area immediately surrounding Louisville, the airport can be reached by taking Preston Highway, which leads to Phillips Lane. Continue west along Phillips Lane and then turn left onto the Freedom Way ramp, and follow signs for the airport.
From Nashville, Louisville International Airport can be reached from Interstate 65 North, leading to Interstate 264 West, exit 131.
From Lexington, Louisville International Airport can be reached from Interstate 64 West, which leads to Interstate 264 West, exit 11.
From St Louis, the airport can be accessed from Interstate 64 East to Interstate 264 East (exit 1) to exit 11.
From Cincinnati, Louisville International Airport can be reached from Interstate 71 South leading to exit 131.
